      Dan Larhammar,研究員,認(rèn)知與衰老研究組組長。瑞典皇家科學(xué)院院士、歐洲科學(xué)院院士、國際科學(xué)理事會會士、烏普薩拉大學(xué)分子細(xì)胞生物學(xué)教授、瑞典皇家科學(xué)院院長(2018-2022)。他于1984年獲得瑞典烏普薩拉大學(xué)醫(yī)學(xué)科學(xué)博士學(xué)位,早期研究集中在對食欲調(diào)節(jié)有重要作用的神經(jīng)肽Y(NPY)的功能。作為訪問學(xué)者在加利福尼亞斯克里普斯研究所工作一年后,他專注于NPY受體的克隆和分子藥理學(xué)研究,發(fā)現(xiàn)了多個(gè)NPY受體,闡明了其在脊椎動(dòng)物中的進(jìn)化過程及藥理學(xué)特性。隨后他的研究擴(kuò)展到了脊椎動(dòng)物眼睛的分子進(jìn)化,特別是視網(wǎng)膜中桿狀細(xì)胞和錐狀細(xì)胞的起源,以及神經(jīng)遞質(zhì)和激素及其受體的進(jìn)化,還研究了與帕金森病相關(guān)的多巴胺受體。近年來,他致力于研究學(xué)習(xí)和長期記憶的分子機(jī)制,重點(diǎn)關(guān)注谷氨酸受體(AMPA和NMDA家族)及其相關(guān)的調(diào)節(jié)蛋白。

認(rèn)知與衰老研究組

本研究組圍繞學(xué)習(xí)和記憶的機(jī)制主要開展兩方面的研究:一方面聚焦于研究谷氨酸受體在學(xué)習(xí)和記憶的過程中的功能以及其與各種調(diào)節(jié)蛋白質(zhì)相互作用的復(fù)雜機(jī)制;另一方面開展對于糖胺聚糖如乙酰硫酸肝素和硫酸軟骨素在學(xué)習(xí)和腦衰老過程中的研究。這些研究工作通過利用轉(zhuǎn)基因斑馬魚、小鼠以及細(xì)胞模型來開展。

相關(guān)的研究方法包括利用單細(xì)胞RNA測序分析識別與長期記憶相關(guān)的細(xì)胞群體和基因,利用空間轉(zhuǎn)錄組學(xué)分析確定學(xué)習(xí)和記憶相關(guān)腦區(qū)表達(dá)的關(guān)鍵分子,利用野生型和轉(zhuǎn)基因型斑馬魚進(jìn)行學(xué)習(xí)和記憶的行為研究,還使用小鼠和斑馬魚模型研究阿爾茨海默病的發(fā)生機(jī)制。
